黑曲霉发酵麦麸生产β-葡萄糖苷酶的培养基组成的优化

摘    要:以黑曲霉为发酵菌株,研究其β-葡萄糖苷酶液体发酵的最佳培养基组成。通过对不同碳源、氮源、磷酸盐、金属离子、氨基酸、表面活性剂等单因素研究,发现麦麸、(NH_4)_2SO_4和KH_2PO_4对产酶有促进作用。3因素二次通用旋转组合设计试验得到该菌株产β-葡萄糖苷酶的优化培养基组成为:麦麸28.4 g/L,(NH_4)_2SO_41.2 g/L,KH_2PO_42.0 g/L,发酵液β-葡萄糖苷酶酶活达到(1 493.90±12.09)U/mL。

Abstract:The optimum media compositions for theβ-glucosidase production from Aspergillus niger are studied.The effects of carbon source,nitrogen source,phosphate source,metal salts,amino acid and surfactant on theβ-glucosidase production are investigated.The results show that wheat bran,(NH_4)_2SO_4 and KH_2PO_4 have stimulatory effect onβ-glucosidase secretion. Theβ-glucosidase activity reaches(1 493.90±12.09) U/mL under the optimum fermentation media based on the three-factor second order rotation combination experimental design.
