羊口疮病毒疫苗株和野毒株F1L基因的比较分析

摘    要:羊口疮(Orf)是由羊口疮病毒(Orf virus,ORFV)引起的人兽共患传染病,ORFV F1L基因编码肝素结合蛋白。为比较分析ORFV疫苗株和野毒株编码的F1L基因的特征,本试验对疫苗株和野毒株的F1L基因进行PCR扩增、克隆及序列测定,并应用生物信息学相关软件及方法,对两者的F1L基因在核酸水平和氨基酸水平的变异情况及其二、三级蛋白结构变化情况进行了比较分析。结果显示,此次测定的ORFV疫苗株与野毒株F1L基因核苷酸序列相似性为95.9%,氨基酸序列相似性为94.5%。本研究结果为揭示ORFV异源宿主致弱的机制积累了资料。

Abstract:Orf is an infectious zoonosis caused by orf virus (ORFV).F1L gene of ORFV encodes a heparin-binding protein.To identify the difference between ORFV vaccine and wild strain,F1L gene was amplified by PCR.The nucleotide and amino acid sequences as well as their secondary and three-dimensional structures were analyzed using bioinformatics software.The results showed that the nucleotide sequence similarity of F1L gene of ORFV vaccine strain was 95.9% to wild strain and amino acid sequence was 94.5%.The results from this study provided useful information for further investigation into ORFV attenuation mechanism by passaging in heterogeneous hosts.
