17份鱼腥草种质亲缘关系的ISSR分析

摘    要:利用ISSR分子标记技术对17份鱼腥草种质进行了基因组DNA多态性分析.从50条引物中筛选出12条多态性引物用于PCR扩增,共扩增出112条DNA条带,其中多态性条带95条,所占比例为84.8%,平均每条引物扩增的DNA条带数目为9.3条.根据ISSR扩增结果,利用POPGENE 1.32软件计算了Nei's基因距离,Nei's基因多样性指数(H)为0.318 5,Shannon信息指数(I)为0.469 9,并用MEGA3.1软件通过UPMGA法进行了聚类分析.结果表明:鱼腥草种质资源具有较丰富的遗传多样性;ISSR技术可将17份供试的鱼腥草材料全部分开,在遗传距离系数0.2处可将17份材料划分成三大类,其中多数材料根据ISSR遗传距离系数划分的类群与地理位置分布有一定关系.同时对鱼腥草道地性与环境因素和遗传因素的关系进行了探讨.

Abstract:17 germplasm resources of Houttuynia cordate were used as materials to analyze their genome polymorphism with ISSR markers.12 primers screened out from 50 primers were used for ISSR amplification.A total of 112 bands were generated,of which 95 bands were polymorphic band(the percentage of polymorphic band,PPB=84.8%).The average number of DNA fragments produced by each primer was 9.3.Nei' similarity coefficients and genetic distances were calculated with POPGENE software and the dendrogram were constructed with UPGMA.17 Houttuynia cordate materials were divided into three major groups while the genetic distance was 0.20.The results indicated that there actually existed much genetic diversity at molecular level among the germplasm resources of Houttuynia cordate,and the genuieness of Houttuynia cordate was related with the environments in some way.The relationship between genuieness,genetic and environmental factors of Houttuynia cordate were discussed.
