| 摘 要： | 应用荧光扩增片段长度多态性（AFLP）标记技术对高邮鸭4个高度纯合的分离体分组混合家系蛋壳颜色性状进行了遗传多样性分析。在高邮鸭分离体分组混合家系AFLP指纹研究中，对AFLP技术中选择性扩增中的引物使用荧光标记代替同位素标记，并对AFLP反应进行了优化。在AFLP分析中，共采用了64个EcoRI／MseI引物组合，每个引物组合扩增出的条带数在40条以上。结果表明：从这64个引物组合中筛选到1个引物组合E-ACT／M—CTC，能同时将供试的4个分离体分组混合家系分开，从而可用于早期青壳性状的鉴定。 |

| Abstract: | This experiment studied AFLP fingerprinting of the bulked extremes of Gaoyou duck. The primers of selecting PCR of AFLP were labeled with fluorescence substituting isotope, experiment technique was optimized, and so the results obtained were excellent. To determine genetic diversity of Gaoyou duck's eggshell colors by means of AFLP labeled with fluorescence among 64 primer pairs. Each pair could give more than 40 amplified fragments. The results showed that the primer pair of E-ACT/M-CTC screened from 64 primer pairs could obviously distinguish the difference band of cyan eggshell and white eggshell trait in the 4 bulked extremes, and it might be used in identifying cyan eggshell trait in the nonage. |
