三花杜鹃组培快繁技术的研究

摘    要:以菌子山采集的优良三花杜鹃单株的茎尖和茎段为外植体,研究三花杜鹃组培快繁技术。结果表明:三花杜鹃组织培养的外植体采用0.1%HgCl2消毒3 min,2%NaClO消毒8min,茎尖的诱导率最高为80%,采用0.1%HgCl2消毒5 min,2%NaC10消毒10 min,茎段的诱导率最高为75%。分别比较了不同培养基对外植体诱导、丛生芽增殖及试管苗生根的效果;初代培养,外植体诱导不定芽最适培养基1/4 MS+ZT 3.0 mg/L+NAA 0.5 mg/L,三花杜鹃茎尖和茎段的诱导率最高,达到90%;增殖培养基采用1/4 MS+ZT 1.0 mg/L+NAA 0.1 mg/L;壮苗培养基采用1/4 MS+ZT 0.5 mg/L+NAA 0.1 mg/L效果最好;三花杜鹃最适生根培养基是1/4 MS+IAA 2.0 mg/L+NAA 2.0 mg/L,生根率为90%。移栽采用泥炭土+珍珠岩+蛭石比例为2∶1∶1作为基质最好,成活率为86%。

Abstract:The tip shoot and stalk node of Rhododendron triflora from Junzi Mountian were used as explants,the tissueculture rapid propagation of Rhododendron triflora were studied.The results showed that the tip shoot on inducing the plant regeneration and the inductivity had reached 80%by 0.1% HgCl2 sterilizing for 3 minutes and 2% NaClO 8 minutes and stalk node had reached 75% by 0.1%HgCl2 for 5 minutes and 2%NaClO for 10 minutes.The comparison of the effects of different media on the explants induction,the prolife...
