摘    要:目的]探讨垂枝樱花外植体灭菌及不定芽诱导的相关技术.方法]以垂枝樱花当年生带腋芽茎段为外植体,对其组织培养繁殖技术中外植体选择、灭菌方法与程序、不定芽的诱导等环节进行了初步研究.结果]适宜外植体采集时间为晴天中午,采集部位为当年生枝条的中上部,适宜的表面灭菌程序为:洗衣粉中浸泡15 min,流水冲洗lh,70%乙醇浸泡30 s,0.1% HgCl2浸泡10 min,最后无菌水冲洗5遍,成活率达68.3%.适宜垂枝樱花不定芽诱导的培养基为1/2MS +6-BA 0.5 mg/L+ NAA 0.10 mg/L,分化率达68.3%.结论]为垂枝樱花的组织培养提供相关技术支持.

The Study on Sterilization of Explant and Formation of Adventitious Bud for Cerasus subhirtella Miq.var.pendula Tanaka.

Abstract:Objective] The aim was to preliminaryly discuss sterilization of explant and formation of adventitious bud for Cerasus subhirtella Miq.var.pendula Tanaka.Method] With the stems with buds of Cerasus subhirtella Miq.var.pendula Tanaka.as explants,the explant selection,methods and procedures of sterilization and adventitious bud induction were studied.Result] The results showed that,the suitable explants acquisition time of midday,collectionsite was in the upper part of annual shoots; the procedures of surface sterilization was immersing 15min of washing powder,water flushing 1 h,70% ethand soaking 30 s,0.1% HgC12 immerising 10 min,the sterile water rinsing 5 times,and reached the survival rate of 66%.The suitable inducing medium was 1/2MS + 6-BA 0.5 mg/L + NAA 0.10 mg/L,and the differentiation rate reached 63.6%.Conclusion] The study provided related technical support for tissue culture of Cerasus subhirtella Miq.var.pendula Tanaka.
