甘蓝型油菜小孢子再生体系的优化研究*

摘    要: 以甘蓝型油菜花油3号、湘油15号为材料,优化了甘蓝型油菜小孢子再生胚状体的条件,建立起甘蓝型油菜小孢子的高效再生体系。在主花序第一朵花开后第4d从主花序上取4.0~4.5mm花蕾,每毫升NLN培养基接种1个花蕾的小孢子,在32℃起始热激培养36h,胚状体再生频率最高,花油3号达24.80个胚状体/花蕾,湘油15号达16.50个胚状体/花蕾。胚状体再生植株的频率为80.25%。

Abstract:Using Brassica napus cv. Huayou 3 and Xiangyou 15 as material, efficient regeneration system of microspore of Brassica napus have been set up by optimum of embryoid regeneration from microspore. Embryoids from microspores were obtained at highest frequency of 24.80 embryoids per flower bud for Huayou 3 and 16.50 embryoids per flower bud for Xiangyou 15 when flowers buds form 4.0mm to 4.5mm were drawn from main inflorescence on the forth day after first flowering, and microspores of a flower bud were inoculated into 1mL NLN culture medium, and heat treatment at the beginning of microspore culture was for 36 hours under 32℃. Plant have been regenerated at frequency of 80.25 percent.
