STS标记检测非糯籼稻蜡质基因的效果研究

摘    要:以82份非糯籼型杂交稻的亲本作为材料,用STS标记Wi10-1和Wi10-2检测其蜡质基因型,并测定其中65份材料的直链淀粉含量,研究STS标记的检测效果。结果表明:STS能准确检测供试材料的蜡质基因型;36份保持系/不育系中30%为WxbWxb,46份恢复系中60%为WxbWxb,16份早稻和早中熟恢复系仅25%为WxbWxb,30份迟熟恢复系中80%为WxbWxb;65份材料直链淀粉含量变幅为11.13%~28.49%,直链淀粉含量与STS标记的相关达1%显著水平(r=0.905);WxaWxa比WxbWxb材料直链淀粉含量平均高9.65个百分点,差异达1%显著水平;同一基因型的材料间直链淀粉含量仍有较大差异,在分子标记辅助选择的基础上,育种材料定选时还需结合直链淀粉含量的直接测定。

Abstract:82 non-glutinous parents of Indica hybrid rice were used as materials to investigate Wx gene by STS molecular markers Wi10-1 and Wi10-2,and the AC of 65 materials among them were investigated to study the test effect of Sequence-Tagged Site(STS) molecular markers.The results showed that STS molecular markers could investigate Wx gene well and truly.30 percentages of 36 maintainer lines and cytoplasm male sterile lines were WxbWxb,60 percentages of 46 restorer lines were WxbWxb,but 25 percentages of 16 early-maturing and middle-maturing restorer lines were WxbWxb,80 percentages of 30 late-maturing restorer lines were WxbWxb.The variation ranges of AC of 65 materials were from 11.13 % to 28.49 %.The correlation between AC and STS molecular markers was significant at 0.01 levels.The average of AC in WxaWxa were 9.65 percent point higher than that in WxbWxb.The differences reached 0.01 significant levels.Meanwhile,there were significant differences of AC among the materials with same Wx genotype;it implied that it was necessary to test the AC directly based on MAS when materials were decided at last during breeding.
