白菜雄性不育相关基因BcMF3和BcMF4的A9启动子的基因沉默植物表达载体的构建

摘    要:以白菜(Brassica campestris ssp.chinensis,syn.B.rapa ssp.chinensis)雄性不育相关基因BcMF3和BcMF4为沉默对象,利用绒毡层组织特异性的A9启动子,构建了反义RNA、RNA干涉和双价反义RNA植物表达载体。结果表明,经过一系列分子操作,获得了A9启动子和目的基因片段,并利用它们构建了5个基因沉默植物表达载体,可用于基因功能验证。

Abstract:Targeting at BcMF3 and BcMF4 genes,which related to male sterility of Chinese cabbage-pak-choi (Brassica campestris ssp.chinensis var.communis,syn.B.rapa ssp.chinensis var.communis),gene silencing vectors of antisense RNA,RNAi (RNA interference) and bivalent antisense RNA were constructed using tapetum-specific A9 promoter.The results indicated that,after a serial molecular manipulation,A9 promoter and target gene fragments were obtained.Subsequently 5 gene silencing plant expression vectors were constructed.These vectors can be used in future functional validation of the two genes.
