杜鹃花组织培养技术研究

试验以东洋杜鹃花 (Rhododendron.L)的茎尖 (含茎段 )、叶片为培养材料 ,研究确定其合适的组织培养各因素条件。结果表明 ,1)由于杜鹃花生长在酸性条件下 ,底盐分浓度及高比值 NH4 + /NO3- 的基本培养基 K和 B适合东洋杜鹃 ;2 )最佳 p H值为 5 .2 ;3)诱导芽的最适外植体为 3～ 5月的顶芽 ,愈伤培养的最适外植体为嫩叶 ;4 )以K& B+2 ,4 - D(1) +6 - BA(0 .0 5 )和 K& B+6 - BA (0 .5 ) +NAA (5 )最适合培养愈伤组织 ,K& B+1AA (1) +2 ,4 - D(4 )对茎尖培养效果较好

Study on the technique system of tissue culture in Rhododendron L.

Studying the suitable condition of culture for tissue culture in Rhododendron L., a experimentation was established in which shoot tip and leaf from different seasons.A series of different medium,sterilization condition and probable way of explant regeneracy,were employed.The results are as follows:1)K&B medium with low mineral salt concentration and high ration of NH 4 +/NO 3 - is suitable for Rhododendron L.callus;2)pH=5.2 is the most suitable;3)The optimal explants are shot tips from Rhododendron L.in 3-5 months for inducing bud.and tender leaf for tissue culture of callus;4)All the cultures are kept under the temperature of 25±2℃,illuminated for 16 hours per-day and light intensity is 1 500 - 2 500 lx;5)The results are showed that K&B medium with 6-BA(0.5)+NAA(5)has the best effect on micropropagation,and the one with IAA(1)+2.4-D(4)has the best effect on inducing bud.
