果梅胚离体培养研究

摘    要:本试验以优选果梅‘黔荔1号’单株成熟胚为材料,对其进行离体培养,并对外植体灭菌时问、胚萌发和增殖以及生根培养进行研究。结果表明:以0.1%HgCl2消毒8min效果最好,其存活率为75.0%;适合胚萌发和增殖的基本培养基为1/2MS+2.0mg/L6-BA+1.0mg/LNAA+3.0mg/LGA3和1/2MS+2.0mg/L6-BA+0.5mg/LNAA+1.0mg/LGA3;适合生根的培养基是1/2MS+0.5mg/LIBA,生根率为91%,平均根数达到3.1。

Research of Prunus mume Embryo Culture in Vitro

Abstract:The optimized single plant (Qian Li NO. 1) from prunus mume embryo as the material was culivated in vitro in order to study explants sterilization, germinating and proliferation and rooting culture of embryo. The results showed that the optimal sterilization time of explant was about 8 minutes with 0.1% HgCI2, and its survival rate was 78.4%. The basal medium for Germination and proliferation were 1/2MS+2.0mg/L6-BA+1.0mg/LNAA +3.0mg/LGA3 and 1/2MS+2.0mg/ L6-BA+0.5mg/LNAA +l.0mg/L GA3. Medium of rooting was 1/2MS+0.5mg/LIBA, and rate of rooting was 91% with average root number of 3.1.
