兰州百合外植体的建立

摘    要:本试验以兰州百合带腋芽的嫩茎为外植体,通过无菌芽诱导、丛生芽增殖及芽苗生根的培养,对兰州百合进行组织培养及快速繁殖技术研究。研究表明:选用6-BA和NAA两种不同浓度的植物生长调节剂对不定芽进行诱导培养,筛选出诱导不定芽的适宜培养基为MS+6-BA1.0mg/L+NAA0.5mg/L,分化率可达88%,平均苗高为3.1cm;选用6-BA和IBA两种不同浓度的植物生长调节剂进行继代增殖培养时,筛选出丛生芽增殖的适宜培养基为MS+IBA0.3mg/L+6-BA0.5mg/L,增殖倍数可达3.4,平均苗高4.6cm;选用IAA和IBA这两种不同浓度的植物生长调节剂进行诱导生根培养,筛选出生根培养的适宜培养基为1/2MS+IAA0.1mg/L+IBA0.2mg/L,生根率90%,平均生根数3.2条。

Establishment of explants of Lilium lanzhouensis
Abstract:The tissue culture and rapid propagation of Lilium Davidii Var. lanzhouensis were studied by inducing aseptic buds, multiplying clustered buds and rooting of buds. The results showed that the optimal medium was MS + 6-BA 1.0 mg/l + Naa 0.5 mg/l, the differentiation rate was 88% , and the average height was 3.1 cm When two plant growth regulators (6-BA and IBA) with different concentrations were selected for subculture, the optimal medium for the multiplication of clustered buds was MS + Iba 0.3 mg/l + 6-BA 0.5 mg/l, the multiplication times could reach 3.4, and the average height of seedlings was 4.6 cm Two different concentrations of plant growth regulators, IAA and IBA, were used to induce rooting culture. The suitable medium for rooting culture was selected as 1/2MS + IAA 0.1 mg/l + Iba 0.2 mg/l, the rooting rate was 90% , and the average rooting number was 3.2. The results showed that the optimum medium for rooting culture was 1/2MS + IAA 0.1 mg/l + Iba 0.2 mg/l.
