摘    要:将LfcinB15-Ma12杂合肽基因克隆到载体pET32a上,构建抗菌肽LfcinB15-Ma12杂合肽基因,并在大肠杆菌中表达融合蛋白。根据已报道的抗菌肽LfcinB和Magainin基因的氨基酸序列,推导出其cDNA序列,将两者连接为杂合基因并克隆到载体pET32a上,IPTG诱导表达。构建了LfcinB15-Ma12杂合肽基因重组质粒,经PCR扩增和DNA测序分析,成功构建了LfcinB15-Ma12杂合肽基因重组质粒,经IPTG诱导,成功表达了杂合肽LfcinB15-Ma12。这为利用基因工程表达其他抗菌肽奠定了基础。

Abstract:In order to get new antibacterial peptide, we designed a hybrid peptide LfcinB15-Ma gainin 12. It synthesized the gene encoding the hybrid peptid. It inserted the gene between the sites of Nco I and Sal I of pET-32a and obtained the recombinant expression vector for heterologous expression of LfcinB15-Ma2 in Escherichia coli. The cDNA sequences were designed based on the amino acid sequences of Lfcin B and Magainin. The hybrid peptide gene was correctly inserted into the vector by PCR amplification and DNA sequencing analysis. It obtained the recombinant plasmid successfully. After induced by isopropyl-β-D-thiogalactoside (IPTG), It realized that the hybrid peptide LfcinB15-Magainin 12 was successfully expressed. This provides a basis for next cost-effective expression of other antimicrobial peptides in genetic engineering.
