棉株中缩合单宁分析条件的优化

摘    要:为探究棉株中缩合单宁的最佳测试提取条件,选取甲醇和乙醇的水溶液作为提取剂,采用香草醛-盐酸比色法测定棉株中缩合单宁的含量。先通过单因素试验确定了甲醇和乙醇各自的最佳提取条件,然后比较这两种提取剂的提取效果,确定乙醇为较好的提取剂,最后通过正交试验确定了乙醇的最优提取条件为:料液比1∶20(g/mL),乙醇浓度40%(V/V),超声时间30 min。

Abstract:This research was designed to discuss the optimal extracting condition of condensed tannin in cotton plant,the tannin was extracted by methanol-water or ethanol-water solvent,then the content of tannin was determined by vanillin colorimetry.Firstly,the best extracting condition of methanol or ethanol was obtained by single-factor experiments.Secondly,ethanol was chosen as a better extracting reagent by comparing the amount of condensed tannin in the best extracting condition of methanol or ethanol.Finally,the optimized extracting condition of ethanol was ascertained by orthogonal experiment,and it showed below: the ratio of material to solvent was 1∶20(g/mL),the concentration of ethanol was 40%(V/V),the time of ultrasonic-extracting was 30 minutes.
