广州香蕉束顶病毒分离和核酸鉴定

摘    要:经液氮冷冻、差速离心和蔗糖梯度离心,从广州地区香蕉束顶病株中获得大小为25nm的病毒颗粒,该病毒与香蕉束顶病毒单克隆兔抗血清呈阳性反应,该病毒核酸可被Dnase和S1核酸酶降解,但不能被RNase降解,属于ssDNA ;人子量大小约为1165碱基。

Abstract:Banana bunchy top disease (BBTD) is the most serious virus-caused disease. The field survey indicated that the incidence of BBTD in South China was 5 %-10 %, even more than 50 % in some areas. In this study, BBTV from Guangzhou areas was purified by way of liquid nitrogen freezing, differential centrifugation, and sucrose density gradient centrifugation, and the BBTV particles of about 25 nm in diameter were obtained. The virus showed a positive reaction with BBTV monoclonal rabbit antiserum. The purified BBTV could be degraded by DNase and S1 nuclease but not by RNase using agarose gel electrophoresis. The results suggested that BBTV was probably a ssDNA virus with a molecular weight of 1165 bases.
