河南省玉米弯孢叶斑病菌遗传多样性的RAPD分析

摘    要:为了解玉米弯孢叶斑病新月弯孢菌的遗传变异情况,利用8个随机引物对来自河南省不同地区的22个新月弯孢菌株进行RAPD分析.结果显示:①22个受试分离株共产生89条谱带,其中70条为多态性带,约占谱带总数的79%;②利用UPGMA法对DNA扩增图谱进行聚类分析,以遗传距离0.65为阈值,将22个受试菌株分为5种遗传类型.上述结果表明,河南省玉米弯孢叶斑病新月弯孢菌群体内具有丰富的遗传多态性,即存在明显的种内遗传分化现象.

Abstract:In order to study genetic variation among Curvularia lunata,genomic fingerprints of 22 isolates of C.lunata isolated from different corn cultivated areas of Hen''an province were analyzed using 8 RAPD primers.The results of RAPD fingerprints showed that there were 89 bands among 22 isolates of C.lunata.Among them,70 bands (79%) belonged to polymorphism bands.The genetic distance and the genetic relationships of the accessions were reconstructed by using UPGMA (Unweigthted Pair-group Method with Arithmetic Average) cluster analysis.All isolates were divided into five genotypes based on threshold 0.65 of genetic distance.All above results indicated that there was abundant genetic diversity among C.lunata colony.
