杨树木质部特异启动子的分离与功能分析

摘    要:从三倍体毛白杨-93(Populus tomentosaCarr.)中分离得到了MDCesAP这一木质部纤维素合酶基因启动子。通过对其进行序列分析,发现A/T含量达63%,有TATA box、polyA等启动子调控元件。与PTCesAP比较,同源性较低。用MDCesAP分别替换PCAMB IA1302及PB I121中的CaMV 35S启动子,转入植物后检测目的片段的启动子功能,结果表明MDCesAP为木质部特异性启动子。

Abstract:The CesA gene promoter MDCesAP was amplified from triploid Chinese white poplar(Populus tomentosa Carr.).Its A/T composition is up to 63%.It has TATA box,polyA and so on regulatory elements in the sequences.The promoter showed lower similarity in the homologous sequences in comparison with the PTCesAP amplified from Populus tremuloids Michx.The MDCesAP was transformed into PCAMBIA1302 and PBI121 for substituting CaMV 35S promoter to study characterization of the promoter,the result indicated that the vectors were introduced into poplar,it was verified that MDCesAP is the xylem specific promoter.
