墨西哥蒲包花愈伤组织诱导及无性系建立的研究

摘    要:以具有有利变异性状的墨西哥蒲包花嫩茎为材料,采用组织培养的方法,对愈伤组织的诱导、分化、不定芽生根、试管苗的生根继代增殖培养,以及试管苗的移栽、定植进行了研究。结果表明:MS+6-BA 0.5 mg/L+2,4-D 3.0 mg/L是嫩茎愈伤组织诱导培养的理想培养基;MS+AgNO3 1.0 mg/L+6-BA 0.8 mg/L+NAA 0.2 mg/L是愈伤组织分化培养的理想培养基;1/3 MS+蔗糖15 g/L+ABT 2.0 mg/L是不定芽生根培养和生根继代增殖培养的理想培养基。试管苗移栽成活率为91.2%,定植成活率为96.8%,定植的试管苗保持了墨西哥蒲包花原有的植物学性状和淡黄色花瓣上呈现出橙色斑点的有利变异性状。

Abstract:In order to meet the demands of seedlings,the tender stems of Calceolaria mexicana which had favorable variation characteristics were used as materials to do the study by using tissue culture methods.The study was mainly on callus induction and differentiation,adventitious buds rooting and rooting of the subculture,and plantlet transplanting colonization.The results showed that MS+ 0.5 mg/L 6-BA + 3.0 mg/L 2,4-D was the optimum medium for inducing the callus from the tender stems;MS+1.0 mg/L AgNO3 +0.8 mg/L 6-BA +0.2 mg/L NAA was the optimum medium for callus differentiation;1/3MS+ 15 g/L sucrose + 2.0 mg/L ABT was the optimum medium for adventitious buds rooting and rooting of the subculture.The survival rate of transplanted tube seedlings was 91.2% and colonization survival rate was as high as 96.8%.The colonization of plantlets maintained not only all biological traits but also the favorable variation characteristics,such as orange spots on pale yellow petals.
