新梨七号茎段组织培养再生植株的研究

摘    要:从植物生长调节物质的不同配比、不同部位诱导不定芽以及炼苗移栽等几个方面对新梨七号离体茎段再生进行研究,获得茎段产生愈伤组织及不定芽再生的适宜培养基,分析以上因素对新梨七号再生体系建立的影响。结果表明,新梨七号的茎段最适宜愈伤诱导培养基为MS+2,4-D 1.0mg/L+6-BA 0.5mg/L,愈伤组织诱导率为100%;茎段诱导不定芽的最佳培养基为MS+6-BA 1.0mg/L+NAA 0.5mg/L,诱导率为95%,增殖倍数为7.29;最适宜不定芽生根培养基为1/2MS+IBA 2.5mg/L+蔗糖15g/L,生根率为90%;封口炼苗时间14d为最好,移栽时间7月最佳。

Abstract:The factors affecting on plant regeneration from stem of pear variety Xinli No.7 in vitro were studied.These factors included ways of stem inoculation,types and concentration of plant growth regulators,explant types,and rooting and transplantation and so on.The result shows that the optimal medium for callus induction was MS + 2,4-D 1.0 mg/L +6-BA 0.5 mg/L,with the callus induction rate of 100%;The optimal medium for plant regeneration was MS supplemented with 6-BA 1.0 mg/L and NAA 0.5 mg/L,regeneration rat...
