发根农杆菌Ri质粒T-DNA对杜仲的遗传转化研究

摘    要:杜仲是中国特有的经济树种,杜仲毛状根能够合成与杜仲含量相当的次生代谢产物。本实验用发根农杆菌ATCC15834分别感染杜仲Eucommia ulmoides Oliv的子叶、叶片、叶柄、下胚轴和根5个营养器官,结果表明杜仲植物的5个营养器官均能单独诱导出大量的毛状根。经PCR检测,发根农杆菌ATCC15834Ri质粒的rolB基因已转入杜仲毛状根基因组中。说明用发根农杆菌诱导杜仲产生大量的毛状根的方法有效,为大规模用发根农杆菌Ri质粒转化到杜仲植物中提供了理论基础。

Abstract:Eucommia ulmoides Oliv was a speicial tree species in China, of which hairy root could synthesize comparative amouut of secondary matabelite. Agrobacterium rhizogenes was used to infect cotyledon ,leaf, petiole, hypocotyl and root of Eucommia ulmoides Oliv in this experiment ,and the result demonstrated that hairy root could be induced from all the five pieces of nutritive organ. The PCR deteoion suggested that rolB of A. rhiwgenes ATCC15834 Ri plasmid was integrated into the genome of hairy roots from Eucommia ulmoides Oliv. It was demonstrated that using Agrobaterium rhizogenes to induce hairy roots of E. ulmoides Oliv was a effective way. The study could provides the theoretical foundation for transforming Ri plasmid of Agrobaterium rhizogenes to E. ulmoides Oliv in large scale.
