陕西杨凌地区番茄斑萎病毒外壳蛋白基因的克隆与分析

摘    要:番茄斑萎病毒(Tomato spotted wilt virus,TSWV)是世界上危害最大的十大病毒之一,为了确定陕西杨凌地区的番茄感染TSWV情况,采集该地区4份疑似感染TSWV的番茄样品,通过表型观察以及PCR分子鉴定进行分析。结果显示,2份材料扩增出234bp的特异性条带,表明样品感染了TSWV。采用同源克隆方法获得了杨凌地区757bpTSWV的外壳蛋白基因(CP),其与西班牙TSWV分离物同源性最高,为99%,而与昆明的TSWV分离物亲缘关系较远。

Abstract:Tomato spotted wilt virus (TSWV) is one of the top 10 harmful viruses in the world.In order to determine the situation of tomato infected by TSWV in Yangling region of Shaanxi,this experiment collected 4 suspected TSWV infected tomato samples from this region,and carried out phenotypic observation and PCR molecular identification.The results showed that 2 samples had amplified a specific band of 234 bp,indicating that they were infected by TSWV.The coat protein gene(CP)of 757 bp TSWV in Yangling region was obtained by homologous cloning method,which had the highest homology with Spanish TSWV isolate,reaching 99%,but had a distant relationship with Kunming TSWV isolate.
