摘    要:目的]运用微滴式数字PCR技术检测转基因玉米品系。方法]基于微滴式数字PCR平台,建立3种转基因玉米品系TC1507、MIR162、MIR604的二重微滴式数字PCR(dd PCR)检测方法。结果]特异性试验结果显示,该法只有特定品系的靶序列才有扩增信号。灵敏度试验表明,该方法在10 pg基因组DNA用量时可定量检测到2~16个拷贝。结论]该研究建立的3种转基因玉米品系定量检测方法特异性强、灵敏度高,可用于进出口农产品中3种转基因玉米品系成分的定量检测。

Study on Detection of Genetically Modified Maize Lines Using Microdrop Digital PCR Technique
Abstract:Objective]To detect the genetically modified maize lines using microdrop digital PCR technique.Method]Based on the droplet digital PCR platform,we established a duplex ddPCR detection method for three kinds of genetically modified maize line TC1507, MIR162, MIR604. Result]The results of specific experiments showed that only the target sequence of a specific strain can be amplified. Sensitivity ex-periments showed that the method can detect 2 -16 copies of 10 pg genome DNA. Conclusion]The quantitative detection methods of the three transgenic maize lines established in this study were highly specific and sensitive,and could be used for the quantitative detection of three GM maize lines components in the import and export agricultural products.
