危害马铃薯的茎线虫分离鉴定

摘    要:本文对采自河北张家口市察北区、危害马铃薯的线虫(编号De-Chabei)进行了形态及分子生物学鉴定,形态测量结果表明其与马铃薯腐烂茎线虫(Ditylenchus destructor Thorne)一致,采用通用引物rDNA1/rDNA2扩增rD-NA-ITS区的长度为1130bp,测序比对以及采用马铃薯茎线虫B型特异性引物DdL1/DdL2扩增得到485bp特异性片段,表明与B型或L型马铃薯腐烂茎线虫一致;再次证明该型线虫在河北马铃薯主产区的分布和危害。

Abstract:A nematode population, named De Chabei, was isolated from potato in the Chabei District of Zhangjiakou City, Hebei Province. It had been identified as type B or L of Ditylenchus destructor Thorne based on morphological and molecular methods. The amplified products of rDNA ITS regions with the universal primers rDNA1/rDNA2 and the specific primers DdL1/DdL2 were 1 130 bp (GenBank accession No. FJ911551) and 485 bp, respectively. The results revealed that this type of D. destructor has existed in the potatogrowing areas of Hebei province.
