双低油菜籽浓缩蛋白制备条件研究

目的]研究更加合理的制备菜籽浓缩蛋白的方法。方法]以乙醇溶液的浓度、pH值、液固比、洗涤次数、洗涤时间为影响因子,设计5因素4水平的正交试验制备浓缩蛋白。结果]结果表明,浓缩蛋白最佳浸提条件为：在55℃水浴中,以pH值为4.5的70%乙醇溶液作为洗涤剂,控制液固比为8：1,洗涤7次,每次20 min。结论]通过这种方法制备的浓缩蛋白中蛋白质的含量高达64.60%,植酸的含量为2.16%,且其中没有检测出单宁。

Study on Preparation of Concentrated Protein from Double-low Rape Seed

Objective]The research aimed to find more better methods for the preparation of rape seed concentrated protein.Method] The L16(45) orthogonal design was used to research the effects of alcohol solution concentration,pH value,liquid-solid ratio,the number of extraction,the time of extraction on the extraction of rape seed concentrated protein.Result]The results showed that the optimum conditions were pH value 4.5 70% alcohol water solution as extraction solvent,extraction temperature 55℃,with ratio of liquid to solid as 8∶1,washing for seven times and 20 min each time.Conclusion]By this way,the ratio of rape seed protein reached 64.60%,the content of phytic acid was 2.16% and the content of tannin was 0.
