摘 要: | [目的]建立不同品种紫花苜蓿的愈伤组织及分化体系。[方法]以4个品种的无菌苗子叶为外植体,研究不同的激素浓度及基因型对紫花苜蓿胚性愈伤组织诱导及分化的影响。[结果]4个品种诱导形成的愈伤组织差别不大;最佳胚性愈伤组织诱导培养基为改良SH+2,4-D 2 mg/L+KT 0.05 mg/L;苜蓿王的分化率和出苗率表现较佳;最佳分化培养基为改良SH+KT 0.4 mg/L。[结论]该试验为今后筛选易分化的苜蓿品种和研究"转基因苜蓿疫苗"奠定了基础。

Abstract: | [Objective] To establish the callus and differentiation system of different alfalfa varieties.[Method] The cotyledon of four varieties of alfalfa(Medicago sativa L.) were used to study the effect of hormones in different concentrations and genotypes on embryogenic callus and differentiation.[Result] The embryogenic callus rate of four varieties of alfalfa were little difference;the best medium for embryogenic callus induction was the modified SH+2.0 mg/L 2,4-D+0.05 mg/L KT;the differentiation and plant development rate of Muxuwang variety was the highest;the best medium for differentiation was the modified SH+0.4 mg/L KT.[Conclusion] The study laid a foundation for screening easy-differentiation alfalfa variety and researching transgenic alfalfa vaccine. |
