摘    要:目的]为了更好地提取真菌黑色素。方法]比较了菌丝粒径(30、60、100和140目)和3种高温提取条件(高压灭菌锅内121℃20 min、沸水浴2 h和沸水浴5 h)提取真菌黑色素的效果。结果]菌丝粒径对黑色素提取效果影响不显著。沸水浴5 h提取不同粒径菌丝黑色素的含量在0.01水平显著高于其余2个高温提取条件。沸水浴5 h提取30目菌丝黑色素含量的RSD为0.5%~3.0%,黑色素加标回收率为86.0%~123.0%。结论]1 mol/L NaOH沸水浴5 h提取30目菌丝中的黑色素是较好的提取方法。

Study of Extracting Condition of Fungal Melanin

Abstract:Objective] The research aimed to extract the fungal melanin better.Method] The effects of the particle size(30,60,100 and 140 mesh) of fungal mycelium and 3 kinds of extraction conditions at high temperature(121 ℃ and 20 min in high-pressure sterilization pot,boiling water bath for 2 h and boiling water bath for 5 h) on the extraction of fungal melanin were studied.Result] No significant influence of the particle size of fungal mycelium on the extraction of fungal melanin was found.The content of fungal melanin was extracted by boiling water bath for 5 h was significantly higher than that by 121 ℃ and 20 min in high-pressure sterilization pot or by boiling water bath for 2 h.RSD and recovery rate of fungal melanin extracted from fungal mycelium of 30 mesh by boiling water bath for 5 h was 0.5%-3.0% and 86.0%-123.0% respectively.Conclusion] The method was feasible for the extraction of fungal melanin from fungal mycelium of 30 mesh by 1 mol/L NaOH with boiling water bath for 5 h.
