PAGE法鉴定西瓜杂交种研究初报

摘    要:本研究利用种子蛋白质聚丙烯酰胺凝胶电泳(PAGE)等多种电泳技术对供试西瓜杂交种及其亲本种子进行了鉴定,结果表明干种子蛋白质PAGE法在分离胶T=14%,C=0.35%,pH5.8;浓缩胶T=4.0%,C=0.33%,pH等于5条件下可将供试西瓜杂交种与其亲本种子明显区分开,因此,这种方法可以作为该组合种子真实性鉴定的手段,作为纯度鉴定方法还需进一步研究。

Abstract:Variety identification has always been the focal and difficult point of seed testing,especially for hybrid water melon seeds. Several kinds of electrophoresis techniques were used in this experiment to identify the hybrid water melon with its marternal and paternal seeds, and results showed that all these 3 varieties tested can be distingnished clearly by utilizing PAGE of dry seed proteins in the condition which T= 14%,C=0.35%, pH5.8 lower gel and T=4%,C=0.33%,pH5 upper gel. So the method can be used to identify the truthfulness of the varieties tested,but much work is being done to apply to genetic purity assay.
