缙云山土壤拮抗放线菌的分离、筛选与发酵条件优化

摘    要:以烟草赤星病菌为指示菌,从缙云山土壤中分离和筛选出6株拮抗放线菌,其中JY-22对指示菌拮抗效果最好。对JY-22的抑菌谱测定结果表明:JY-22对棉花枯萎病菌、烟草赤星病菌、小麦纹枯病菌、柑橘疮痂病菌、茶云纹叶枯病菌和柑橘青霉病菌都具有较强的拮抗作用,抑菌率达41.9%~75.7%。通过单因子和正交试验对拮抗放线菌JY-22进行发酵条件的研究,结果表明,其最佳培养基组成和培养条件为:1 000 mL培养基中含葡萄糖2.0%、玉米粉1.5%、黄豆粉3.0%、氯化钠0.25%、碳酸钙0.2%,初始pH值为自然,250 mL三角瓶装液量为50 mL,发酵周期为72 h。

Abstract:Six antagonistic isolates of Actinomycetes spp.were isolated from the cultivated soil samples of Jinyun mountain,among which JY-22 exhibited the highest antagonistic activityagainst Alternaria alternate.Further antimicrobial spectrum of JY-22 was tested against Alternaria alternate,Bipolaris maydis,Fusarium oxysporum,Ceratobasidium cornigerum,Sphaceloma fewcetti,Colletotrichum cameliae and Penicillium italicum.The results indicated that JY-22 showed the inhibition rate between 41.9% to 75.7 %.The optimal fermentation conditions for antibacterial substance production of JY-22 were pH being natural,28℃ in yeast peptone glucose liquid medium shaking for 72 h with 50mL fermentation fluid in a 250mL triangular flask.
