福建平潭月见草组织培养与快繁技术

摘    要:以白天开花的平潭月见草种子为外植体,进行月见草的组织培养与快繁技术研究。结果表明,选择成熟度较高的种荚,在无菌条件下取出种子,用75%酒精处理30 s,然后用0.1%升汞灭菌15 min,最后用无菌水荡洗5遍,能达到较好的灭菌效果;用滴管滴植后,再用镊子栽植的接种方式较佳;最适宜的试管苗增殖培养基为MS+2.0 mg.L-16-BA+0.1 mg.L-1NAA固体培养基;壮苗生根培养基以固体培养基1/2 MS+0.1 mg.L-1NAA为最佳培养基;蛭石∶泥炭土∶河沙=1∶2∶1的混合基质为最佳移栽基质,成活率可达93.33%。

Abstract:The seeds of Oenothera biennis L.flowering during daytime and collected from Pingtan,Fujian were used as explants to study its tissue culture and rapid propagation.The treatment and results showed that under sterile conditions,the seeds from pods of high maturity were soaked in 75% alcohol for 30 seconds,and 0.1% HgCl2 for 15 minutes,and rinsed by asepsis water for 5 times,and optimal sterilization was achieved.The favorable planting method was dropping with the dropper,then planting with tweezers.The solid medium of MS medium supplemented with 2.0 mg·L-1 6-BA and 0.1 mg·L-1 NAA was suitable for multiplication of shoots.The solid medium of 1/2 MS medium supplemented with 0.1 mg·L-1 NAA was best for growth and rooting.The best mixture matrix for transplanting,vermiculite∶ peat∶ sand(1∶ 2∶ 1),improved the rate of survival,reaching as high as 93.33%.
