满天星茎尖试管苗的继代培养基研究

摘    要:
探讨了满天星试管苗继代培养过程中,不同生长调节剂、碳源、水质、活性碳对芽增殖效果的影响.结果表明,NAA 0.5 mg/L+6-BA 1.0 mg/L的生长调节剂组合下,试管苗增殖效果最好,芽的长势好,增殖倍数达5.0;用40 g/L的市售白糖代替30 g/L的化学纯蔗糖作为培养基的碳源,既可降低配制培养基的成本,又不影响苗的增殖和质量,可用于满天星试管苗的商业化生产;但以自来水代替蒸馏水对试管苗进行继代培养,则不利于苗的生长和增殖;在培养基中加入0.2%的活性碳有利于试管苗株高的增加,但不利于芽的增殖.

Optimization of subculture media of test-tube shoots from stem apexes of Gypsophilo paniculata

Abstract:
The effects of different growth regulators,carbon source,water quality and 0.2 % active carbon in culture media on bud proliferation in the subculture of test-tube shoots from stem apexes of Gypsophila paniculata were studied.The results showed that the optimum combination of growth regulators in the culture media for bud proliferation of test-tube shoots was NAA 0.5 mg/L coupled with 6-BA 1.0 mg/L,in which proliferation buds had better growth vigor and proliferation multiples of adventitious buds reached to 5.0.Commercial white sugar 40 g/L could took the place of chemical pure cane sugar 30 g/L and was used as the carbon source of culture medium,so as to decrease the preparation cost of culture medium as well as keep the proliferation and quality of test-tube shoots,thus could be applied to commercial production of test-tube shoots of Gypsophila paniculata.But tap water could not replace distilled water for the subculture medium because it was unfavourable for bud proliferation and growth of test-tube shoots.Adding 0.2 % activated charcoal into the medium could promote elongation of test-tube shoots but inhibit bud proliferation.
