| 摘 要： | 为建立PCV1和PCV2混合感染快速检测的PCR方法,根据GenBank已发表PCV1和PCV2全基因组序列,设计合成4条引物,通过PCV1和PCV2阳性质粒的构建、反应条件的优化、敏感性试验和特异性试验,建立了PCV多重PCR检测方法,可扩增出PCV1和PCV2长度分别为726 bp和433 bp特异性目的基因片段。结果显示,建立的PCV多重PCR可检测到5×101个拷贝的PCV1和PCV2目的基因,HCV、PRV、PPV核酸扩增均为阴性,具有良好的特异性和敏感性。并初步应用建立的方法对42份采自四川省部分地区的样品进行检测,结果表明,PCV1阳性率为33.3%,PCV2阳性率为45.2%,其中PCV1和PCV2混合感染阳性率为16.6%。 |
