草地贪夜蛾肠道微生物宏基因组初步分析

摘    要:为了解草地贪夜蛾Spodoptera frugiperda暴发机制,探索新型绿色防控策略,通过构建16S rRNA文库和宏基因组文库系统解析草地贪夜蛾5龄幼虫肠道微生物的特征及功能。结果显示,16S rRNA文库主要鉴定细菌种群,共鉴定4门50属,其中厚壁菌门和魏斯氏菌属分别为草地贪夜蛾幼虫肠道优势门和优势属,其平均相对丰度分别为99.715%和72.046%。宏基因组分析鉴定到的微生物类群更丰富,除细菌外,还检测到真菌、病毒及少量古细菌,其平均相对丰度分别为84.227%、3.099%、2.564%和0.004%;共鉴定到的细菌有54门605属,厚壁菌门和魏斯氏菌属分别为草地贪夜蛾幼虫肠道优势门和优势属,平均相对丰度分别为81.236%和38.247%,与16S rRNA文库鉴定的菌群种类和丰度一致。草地贪夜蛾幼虫肠道微生物碳水化合物代谢和膜运输通路的丰度最高,表明草地贪夜蛾幼虫肠道微生物在肠道内发挥较强的代谢功能。

Abstract:To characterize the gut microorganisms and their functions of fall armyworm Spodoptera frugiperda, the 16S rRNA library and metagenomic library were constructed from 5th instar larvae of S. frugiperda. The results of metagenomic analysis of the 16S rRNA library showed that the gut microorganisms mainly included bacterial populations in four phyla and 50 genera. The dominant phylum in the intestinal tract of S. frugiperda larvae was Firmicutes, and Weissella was the main dominant genus, with an average relative abundance of 99.715% and 72.064%, respectively. The microbial taxa identified by metagenomic analysis were more abundant, and in addition to bacteria, fungi, viruses and a small number of archaea were also detected, with average relative abundances of 84.227%, 3.099%, 2.564% and 0.004%, respectively; a total of 54 phyla and 605 genera were identified, with the dominant phylum being Firmicutes, in which Weissella was the dominant genus. The relative average abundance was 81.236% and 38.247%, which was consistent with the species and abundance of the bacterial groups identified from the 16S rRNA library. The functions identified from metagenomics analysis with high abundance were carbohydrate metabolism and membrane transport pathway, indicating that the gut microorganisms of S. frugiperda larvae play an important role in metabolic function of the intestine.
