摘    要:采用RT-PCR对3株H9N2亚型禽流感病毒基因组进行扩增,测序后再利用DNAStar和MEGA 4对所得到的序列和部分具有代表性的参考序列进行遗传演化及分子特点分析,旨在从分子水平探讨华南地区H9N2亚型禽流感病毒的流行特点,为该病的综合防控提供依据。结果表明,3个分离株CK/GD/162/03、Francolin/GD/298/05与CK/GD/A8/10分别属于B34、B47和B54基因型;3个分离株HA基因未发生明显的变异,符合我国大陆H9N2亚型AIV HA基因的遗传进化特点,但内部基因出现不同程度重组且部分基因与人源高致病性H5N1亚型AIV同源性很高;CK/GD/162/03、Francolin/GD/298/05两株分离株具有宿主多样性。因此,对华南地区H9N2亚型AIV进行监控及分子流行病学调查具有重要意义。

Abstract:Full length cDNAs of the whole genome of three avian influenza virus strains were amplified by RT-PCR and were sequenced.Then,DNA Star and MEGA4 were used to study the phylogenetic and molecular characterization of H9N2 AIV in south China in order to provide the deta support for this virus prevention.The results showed that CK/GD/162/03,Francolin/GD/298/05 and CK/GD/A8/10 belonged to B34,B47and B54 subtype.It was shown that these viruses exhibited the common feature of H9 subtype AIV from China,but the internal genes were reassorted,and two viruses(CK/GD/162/03,Francolin/GD/298/05) owed many hosts.Therefore,the continuing evolution of H9N2 influenza viruses emphasizes the importance of H9N2 influenza virus surveillance and molecular epidemiology investigation in south China.
