棉花黄萎病菌拮抗细菌TYG-2抑菌蛋白的分离纯化

摘    要:
拮抗细菌TYG-2是从根际土壤中分离得到的一株芽孢杆菌,其菌株发酵液产生的抑菌物质对棉花黄萎菌具有较强的抑制作用。为了得到它的抑菌活性组分,将TYG-2菌株去菌体发酵液经硫酸铵沉淀,得到的沉淀物脱盐后经DEAE-Sepharose Fast Flow分离得到4个组分,经抑菌活性测定得到一个抑菌活性组分,再通过Sephacry S-100凝胶过滤分离得到两个组分P1和P2,经检测表明P1组分对V.dahliae的菌丝生长具有抑制作用,为有效抑菌活性组分。经SDS-PAGE电泳纯化为单一蛋白带,分子量约为30kDa。

Abstract:
Bacillus TYG-2was isolated from the cotton soil,whose fermentation liquid had antifungal substance against Verticillium dahilae Kleb.In order to get the antifungal substance,precipitates of the fermentation liquid from strain TYG-2 were collected by saturation ammonium sulfate.When the antifungal substance was purified by chromatography after desalting,four peaks were obtained from DEAE-Sepharose Fast the Flow,the fourth peak showed antifungal activity.After purified through Sephacry S-100,one of the two pe...
