黑木耳菌丝深层培养的工艺及其应用

摘    要:以Au.793菌株为材料,研究了8种营养物质对菌丝生长的影响,并通过正交试验得到深层培养黑木耳菌丝的适宜培养基配方:葡萄糖0.5%,麸皮4%,黄豆粉0.2%。同时确定了深层培养黑木耳液体菌种的工艺条件:温度28℃,培养基pH值6~6.5,罐压0.6kg/cm~2,通气量0.25~0.5v/v/m,搅拌速度500rpm,接种量10~15%,种龄48小时。用液体菌种进行代料栽培,结果表明:液体菌种比传统的固体菌种迟缓期短,污染率低,生长快,可提前10~12天出耳。

Abstract:Some natural materials used as nutritional sources for the mycelial growthof Auricularia auricula were studied in the present paper. A suitable liquid mediumformulation for the submerged culture was proposed, according to the results of certainpreliminary tests of the research. Results of the effects of temperature, pH, shakingspeed, volume of the medium, viscosity of the medium, aeration, stirring speed, and theage, together with the amount of the inoculum on the mycelial growth and itsmorphological character in submerged culture were presented. A promising submergedculture technique was recommended. The experimental results showed that the liquid spawnhas several advantages over ther the solid spawn, such as shortening the time ofgermination, reducing incidences in contamination, increasing the growth speed andshortening the duration of cultivation, and fruiting ahead of schedule.
