摘 要: | 为明确引起福建省福州市长乐地区番茄上曲叶症状的病原,提取样品总DNA,利用简并引物PA/PB进行PCR扩增,从10个样品中均可扩增到500 bp左右的特异条带.NCBI BLAST序列比对表明,该序列与番茄黄化曲叶病毒(TYLCV)核苷酸序列相似性最高,达99.0%.再根据已知500 bp序列设计1对反向引物,扩增TYLCV分离物Fz01全基因组近全长,克隆并测序.经序列拼接发现,TYLCV分离物Fz01基因组全长2781 nts(KP685598),与已报道的TYLCV其他各分离物相似性均在89.3%以上,而与来自中国不同地区的TYLCV分离物的相似性均在98.7%以上.因此,Fz01属于TYLCV的一个分离物.

Molecular characterization of Tomato yellow leaf curl virus in Fuzhou |

Abstract: | To identify pathogens that caused tomato leaf curl symptoms in Changle area of Fuzhou, Fujian Province, total DNA was extracted from 10 infected samples. By using degenerate primers PA/PB, 500 bp fragments were obtained from all samples. Se-quence analysis showed nucleotide sequences of all fragments showed the highest identity (99.0%) with that of Tomato yellow leaf curl virus ( TYLCV) . To get the complete nucleotide sequence of virus isolate Fz01, a pair of reverse primers was designed according to known genome sequence. Results showed that isolate Fz01 comprised of 2781 nucleotides, reaching 89.3% identity with other iso-lates of TYLCV from GenBank, and more than 98.7% identity with other isolates of TYLCV from China. Therefore, the virus that in-fected tomato in Fuzhou was an isolate of TYLCV. |
