超声波辅助提取长梗黄精多糖工艺的研究

摘    要:以梅列产的长梗黄精为原料,研究液料比、提取温度、超声波功率和超声波时间等因素对超声波辅助提取长梗黄精多糖得率的影响规律,经优化确定超声波提取长梗黄精多糖的最佳工艺。由单因素试验结果确定液料比、提取温度、超声波作用功率和超声波处理时间各因素的优化参数,采用DPS 9.05和Design Expert7.0数据分析软件,建立响应面模型进行结果分析,通过响应面法分析结果获得各因素与长梗黄精多糖得率的交互关系,从而确定超声波提取长梗黄精多糖的最佳工艺条件。结果显示长梗黄精多糖优化后的提取工艺为:液料比18:1,超声波处理时间59.7min,浸提的温度72.9℃,超声波作用功率152.8W,所得的长梗黄精多糖的得率理论值16.65%。经响应面模型验证,实际得率为16.59%,表明该模型可靠。

Ultrasound-assisted Extraction of Polysaccharides from Polygonatum Filipes
Abstract:Effect of various factors in an ultrasound-assisted process on the polysaccharide extraction from Polygonatum filipes were studied for the technology optimization.The liquid/material ratio,temperature,as well as power and duration of ultrasonic application for the process were scrutinized.A quadratic regression rotational combinational design and the Design Expert 7.0 analytical procedures were applied to establish a regression model for projecting the maximal extraction yield under optimized conditions.Theoretically,the optimized process included the application of a liquid/material ratio of 1 7.81 mL/1 g at 72.97℃ with a 1 52.85 W ultrasonic treatment for 59.74 min to reach 1 6.6471% yield on polysaccharides.In a triplicated test run,an averaged polysaccharide yield of 1 6.59% was obtained with a liquid/material ratio of 18 mL/1 g at 73℃ with a 1 50 W ultrasonic treatment for 60 min.The result showed that the mathematic model was accurate in predicting the performance of the extraction process.
