苇状羊茅EST-SSR标记在草甸羊茅和多年生黑麦草的通用分析

摘    要:草甸羊茅(Festuca pratensis Huds)和多年生黑麦草(Lolium perenne Lam)均是禾本科、多年生草本植物,是优良的牧草和草坪草。本研究采用40对苇状羊茅(Festuca arundinacea Schred)的表达序列标签-简单序列重复(Expressed sequence tags-simple sequence repeats,EST-SSR)分子标记分别评比了18份草甸羊茅和18份多年生黑麦草的遗传的多样性,并发现上述标记在草甸羊茅和多年生黑麦草中的通用性比例分别为55%和50%。在草甸羊茅中22对引物共扩增出212个条带,每对引物扩增条带数在4~13之间,多态性条带总数为190条,百分率为89.62%;多年生黑麦草中20对引物共扩增出203个条带,条带数在4~16之间,多态性条带总数为182条,百分率为89.67%;另外,分子标记NFA103和NFA140可以直接用于区别多年生黑麦草和草甸羊茅。综上,本研究发掘的通用性引物对于两种草的遗传多样性分析及种质资源合理利用具有重要的意义。

Abstract:Meadow fescue (Festuca pratensis Huds) and Perennial ryegrass (Lolium perenne Lam) are both grass and perennial herb, they are excellent forage and lawn grass. Forty pairs of EST-SSR markers of tall fescue(Festuca arundinacea Schred)were used to evaluate the genetic diversity of 18 meadow fescue cultivars and 18 perennial ryegrass cultivars. The result showed that these markers were 55% and 50% common in meadow fescue and perennial ryegrass. A total of 212 bands were amplified by 22 pairs of primers, and the number of bands amplified by each pair of primers ranged from 4 to 13. The total number of polymorphic bands was 190, with a percentage of 89.62%. A total of 203 bands were amplified by 20 pairs of primers from perennial ryegrass, and the number of bands ranged from 4 to 16. The total number of polymorphic bands was 182, with a percentage of 89.67%. In addition, molecular markers NFA103 and NFA140 can be directly used to distinguish perennial ryegrass from meadow fescue. In conclusion, the general-purpose primers unearthed in this study are of great significance for the analysis of genetic diversity and rational utilization of germplasm resources of the two kinds of grass.
