塔里木盆地一处柽柳林土壤细菌多样性分析

摘    要:【目的】对塔里木盆地一处柽柳林土壤细菌多样性进行初步探索,为下一步对塔里木盆地其它地区柽柳林土壤微生物多样性分析及极端环境微生物的筛选奠定基础。【方法】采集塔里木盆地一处柽柳林土壤样品,采用纯培养方法进行细菌的分离纯化。提取各菌株基因组DNA,进行16S rRNA扩增、测序及系统进化树的绘制,分析其多样性。【结果】共分离得到21株菌,其中Firmicutes(厚壁菌门)11株,Actinobacteria(放线菌门)7株,Proteobacteria(变形菌门)和Bacteroidetes(拟杆菌门)各1株。革兰氏染色结果表明,3株菌为革兰氏阴性,其余为革兰氏阳性。Blast比对结果表明菌株CL4与标准菌株Pontibacter korlensis(DQ888330,普拉特链霉菌)的同源性为91%,可能为潜在的新种。【结论】塔里木盆地柽柳林土壤中存在较丰富的细菌资源,并可能存在微生物新物种,具有进一步研究开发的价值。

Abstract:【Objective】To investigate the diversity of bacteria in the soil of one Tamarix ramosisima Forest in Tarim Basin and establish an isolation method for the extremophiles.【Method】Soil of one Tamarix ramosisima Forest in Tarim basin was sampled and culture-dependent method was used to isolate the bacteria.After that,gram staining was performed.In order to analyze the phylogenetic relationships of these microorganisms,genomic DNA of each strain was extracted and PCR was carried out with 16S rRNA gene primers(27F and 1492R).Then the 16S rRNA gene fragments were sequenced and the phylogenetic tree was constructed.【Result】A total of 21 strains were obtained from the soil of Tamarix forest in Tarim basin.Among these strains,11 strains were classified as Firmicutes,seven strains were Actinobacteria,one strain was Proteobacteria and one was Bacteroidetes.Gram staining indicated that three strains were gram-negative and the others were gram-positive.16S rRNA gene sequence analysis showed that the homology between the isolates CL4 and the type strain Pontibacter korlensis(DQ888330) was 91% and the strain CL4 was considered to be a candidate novel species.【Conclusion】The bacterial population diversity is abundant in the soil of Tamarix ramosisima Forest in Tarim basin,and there might be novel species exist in the area,which is worth for further research and development.
