无机营养元素对小麦秸秆降解菌降解效果的影响

摘    要:为了研究不同无机营养元素对秸秆降解菌株降解效果的影响,对8种大量元素和微量元素的无机盐进行单因素、多因素组合摇瓶试验,以确定2种降解菌降解小麦秸秆的最佳无机营养条件,并进一步对其在不同pH和时间内的降解效果进行探究.结果表明:菌株DG75B的最佳无机营养元素及用量为Ca 0.005%、Mg 0.015%、Mn 0.0005%、Fe 0.0005%、M0 0.0005%,在最佳无机营养条件培养下,接种处理5天小麦秸秆失重率达到40.59%,比对照CK高10.29%,最适降解秸秆的pH 6~7左右.菌株LYZ22的最佳无机营养及用量为Ca 0.01%、Mg 0.03%,最适pH 6左右,摇瓶条件下,秸秆失重率及其各组分变化均为先缓慢后加速,在40天内处理TEstrain全秸秆失重率达到43.19%,比Tstrain提高了16.07%,秸秆纤维素、半纤维素和木质素降解率分别是43.18%、54.77%、22.96%,与Tstrain相比分别提高了17.29%、15.36%、11.28%.盆钵条件下,秸秆失重率及其各组分变化为先快后慢,60天内TEstrain全秸秆失重率为48.48%,比Tstrain提高21.88%,秸秆纤维素、半纤维素和木质素降解率分别是48.27%、51.41%、13.83%,与Tstrain相比分别提高了17.95%、26.52%、15.81%.

Abstract:

In order to study the effects of different inorganic elements to biodegradation strains, eight macro elements and trace elements were analyzed by univariate and multivariate analysis in shake flask, thus the optimum nutritional conditions that two biodegradation strains degraded wheat-straw were determined, and then the degradation effects during different pH and time were discussed. Results indicated that, the optimum inorganic nutritional condition and dosage of DG75B strain were Ca 0.005%, Mg 0.015%, Mn 0.0005%, Fe 0.0005%, Mo 0.0005%, on this condition, wheat-straw weightlessness rate was 40.59% after 5 days’ treatment, increased by 10.29% in comparison with the control. And the optimum pH to straw degradation was about 6~7. The optimum nutritional condition and dosage of LYZ22 strain were Ca 0.01%, Mg 0.03%, and the optimum pH was about 6. During forty days, variation trend of wheat-straw weightlessness rates and different component contents were from slow to fast in flask, straw weightlessness rate was 43.19%, increased by 16.07% compared to Tstrain. The contents of straw cellulose, hemicellulose, lignin decreased by 43.18%, 54.77%, 22.96%, in comparison with Tstrain the rates increased by 17.29%, 15.36%, 11.28% respectively. Trend of wheat-straw weightlessness rates and different component contents were from fast to slow in experiment of straw returning in sixty days; straw weightlessness rate was 48.48%, increased by 21.88% compared to Tstrain. The contents of straw cellulose, hemicellulose, lignin decreased by 48.27%, 51.41%, 13.83%, the rates respectively increased by 17.95%, 26.52%, 15.81% in comparison with Tstrain.
