卡片PCR法检测果汁饮料中卡耶他环孢子虫技术的建立

摘    要:为建立一种快速检测果汁饮料中卡耶他环孢子虫的方法,采用FTA卡片法提取果汁饮料中的卡耶他环孢子虫DNA,利用特异性引物组F1E/R2B、CC719/CRP999进行巢式PCR。以柔嫩艾美耳球虫、贝氏隐孢子虫等作为对照,结果显示,除卡耶他环孢子虫外,均无特异性条带。用卡耶他环孢子虫卵囊进行巢式PCR敏感性试验,结果显示敏感度最高达1.0×10^0个/mL。可见,FTA.PCR方特异、敏感,能用于新鲜食品中卡耶他环孢子虫的检测。

Abstract:A 298-bp region of the small subunit rRNA gene was extracted from Cyclospora cayetanensis in juice by FTA and amplified in a nested PCR using unique primers ofF 1E/R2B and CC719/CRP999. The amplified product was specific for C. cayetanensis. No DNA amplification occurred when DNA templates from Eimeria tenella and C. baileyi were used. The sensitivity was 1.0×10^2 oocyst/mL. The FTA-PCR assay appears to be specific and sensitive and can be used for the detection of C. cayetanensis in fresh food.
