摘    要:
对三叶崖爬藤ISSR-PCR扩增反应体系的建立和优化进行探讨。结果表明,最适宜的PCR反应条件为20μLPCR反应体积中含有1×缓冲液,20 ng模板DNA,3.0 mmol/L MgCl2,0.2 mmol/L dNTPs,1.5 U Taq DNA聚合酶,0.5μmol/L引物。扩增程序为:94℃预变性5 min;再35个循环:94℃60 s,52℃60 s和72℃105 s;最后72℃延伸10 min。

Abstract:
Factors which affect the ISSR analysis in the molecular study of Tetrastigma hemsleyanum were studied for optimizing conditions.The results showed that the conditions being suitable for ISSR-PCR of T.hemsleyanum were as follows: 1 buffer,20 ng template DNA,3.0 mmol/L MgCl2,0.2 mmol/L dNTPs,1.5U Taq DNA polymerase,0.5 μmol/L primer.The amplification cycling was 1 cycle at 94 ℃ for 5 min;35 cycles at 94 ℃ for 60 sec,52 ℃ for 60 sec,and 72 ℃ for 105 s,respectively,and 1 cycle at 72 ℃ for 10 min.
