香蕉未成熟雄花组织培养与快速繁殖研究

摘    要:
以香蕉未成熟雄花为外植体,进行组织培养和快速繁殖研究。结果表明,香蕉未成熟雄花外植体不消毒处理,在离体24 h内切片厚度1.2 mm为最佳外植体处理方式。最佳愈伤组织诱导培养基为MS+TDZ 0.2 mg/L+Zeatin 0.2 mg/L+生物素1.0 mg/L+谷氨酰胺100 mg/L+糖40 g/L(pH值5.3);最佳胚性分化培养基为MS+6-BA 4.0 mg/L+NAA 0.3 mg/L+糖30 g/L(pH值5.8);最佳生根培养基为MS+IBA 2.0 mg/L+NAA 0.2 mg/L+蔗糖30 g/L(pH值5.8)。炼苗后移栽至椰糠基质中,移栽成活率100%。

Abstract:
The banana immature male flowers were used as the explants for tissue culture and rapid propagation in this study.The results indicated that the best way for explant processing was that the immature male flowers were sectioned as 1.2 mm thickness within 24 hours without disinfects processing.The results also indicated that the best inducing medium for callus induction was MS medium with 0.2 mg/L TDZ,0.2 mg/L Zeatin,1.0 mg/L biotin,100 mg/L glutamine,40 g/L sugar(pH5.3).The best medium for bud differentiation was MS medium with 4.0 mg/L 6BA,0.3 mg/L NAA,30 g/L sugar(pH5.8).The best medium for root differentiation was MS medium with 2.0 mg/L IBA and 0.2 mg/L NAA,30 g/L sugar(pH5.8).After acclimatization,the culture-bottle seedlings were transplanted into the base materials of coconut tree branny.The survival rate was up to 100%.The reproduction technology improved the efficiency of embryo callus induced by banana callus and banana propagation coefficient,providing prerequisite for banana genetic improvement and large-scale production.
