培养基、蔗糖和激素对苜蓿花药愈伤组织诱导的影响

摘    要:为了提高苜蓿花药培养愈伤组织诱导率,以小孢子发育处于单核赤边期中期到单核靠边期的花药为材料,对不同基本培养基、蔗糖浓度和激素配比对花药愈伤组织诱导的影响进行了研究.结果表明,以改良Mitsch2为基本培养基,附加2,4-2、6-BA 0.5 mg/L、蔗糖6;~9;,可以获得较高的诱导率.培养基中的蔗糖浓度和激素配比影响愈伤组织的生长速度和褐化率.

Abstract:In order to improve the anther cultivation of alfalfa, effects of some factors on the callus induCtion rate in anther culture were studied. These factors are basic medium, concentration of sucrose and exogenous hormones. Alfalfa anthers were used as explants, in which pollen was in the middle development stage and/or in the side development stage of single nuclens. The results indicated that induction rate is better with improved Nitsch2 medium, supplemented with 2,4 - D 2 mg/L,6 BA 0.5 mg/L and sucrose 3% - 6%. Concentrations of sucrose and exogenous hormones had effects on the growth rate and browning rate of callus.
