不同培养条件水滴伪康纤虫种群生长研究

摘    要:为揭示水滴伪康纤虫在不同培养条件下的种群动力学规律,初步探讨不同温度(6、12、20℃)及不同培养液条件(虾肉浸出液、麦粒浸出液、牛肉浸膏培养液和蛋白胨培养液)对其种群增长的影响。结果显示,4种培养液在设定浓度范围内,水滴伪康纤虫的种群密度呈现随着食物浓度增大而增长的趋势;不同温度下,6℃时繁殖速度慢,经历长时间(9 d左右)的停滞期后进入增长期,各培养液组在第12 d时达到种群密度峰值,但种群密度低;20℃时各培养液组虫体繁殖快,牛肉膏培养液组在第3 d 即达到种群密度的峰值(57490 ind · mL -1),但衰退迅速;12℃时,4种培养液中的种群密度均显著高于其他2个温度处理,且稳定期维持时间长,更适宜水滴伪康纤虫种群生长。12℃时各培养液的最大种群密度表现为:牛肉膏(2.25 g·L -1)>虾肉(4.5g·L -1)>麦粒(10.14g·L -1)>蛋白胨(2.25g·L -1);种群自然增长率高低表现为:牛肉膏(2.25 g·L -1)>麦粒(10.14g·L -1)>虾肉(4.5g·L -1)>蛋白胨(2.25g·L -1),虾肉与麦粒浸出液种群密度能达到较大值,且稳定期明显长于其他培养液。结果表明,牛肉浸膏培养液适用于短期种群的快速扩大培养,虾肉及麦粒浸出液适用于试验室保种,水滴伪康纤虫在12℃培养可达到种群密度的最大值,20℃时种群自然增长率最高。研究结果提供了试验室培养和保存水滴伪康纤虫的方法,并为海水养殖动物的水滴伪康纤虫病研究提供了试验生态学数据。

Effects of Different Culture Conditions on the Population Growth of Pseudocohnilembus Persalinus

Abstract:Three different temperatures (6 ,12 and 20℃) and four culture mediums (lixivium of shrimp flesh , lixivium of wheat , beef extract and peptone culture solution ) were used to reveal the population dynamics of P .persalinus in different conditions .The results showed that :the proliferation rate of the population density at 6℃is slow ,increased with the concentration of culture media ,reached growth period at day 9 and got the maximum on day 12 ;at 20℃ , the population density increased rapidly and the group of beef extract reached the maximum population density (57 490 ind · mL -1 ) on day 3 ;at 12℃ ,the population density showed :beef extract〉 wheat lixivium 〉 shrimp flesh lixivium〉 peptone .The largest population density of the optimal concentration of culture solution at 12℃ showed :beef extract (2.25 g · L -1 ) 〉 shrimp flesh (4.5 g · L -1 ) 〉 wheat (10.14 g · L -1 )〉 peptone (2.25 g · L -1 );the population growth rates showed :beef extract (2.25 g · L -1 ) 〉 wheat (10.14 g · L -1 ) 〉 shrimp flesh (4.5 g · L -1 ) 〉 peptone (2.25 g · L -1 ) ,therefore ,in lixivium of shrimp flesh and wheat the population density was bigger and then the stationary phase was significantly longer than other culture solutions .Our results suggested that beef extract conld be applied to rapidly expanding growth of short-term population ,the lixivium of shrimp flesh and wheat conld be applied to laboratory breeds conservation ,and the population density and population growth rate could reach the maximum at 12℃ and 20℃ ,respectively .These findings will be benefit for the providing of a laboratory culture and preservation method of P .persalinus ,and provide experimental ecological data of P .persalinus in marine aquaculture also .
