老年大鼠海马CA1区锥体细胞凋亡的形态学观察

摘    要:采用股动脉放血并双侧颈总动脉夹闭法制作全脑缺血再灌注模型,于术后3d处死动物取脑,以常规电镜程序制作标本,观察海马CA1区锥体细胞超微结构的改变;同时结合原位末端标记法检测,探讨全脑缺血再灌注后海马CA1区锥体细胞的病理损伤形式。结果表明:与假手术相比.缺血再灌注3d后海马CA1区出现了较多的凋亡样改变的锥体细胞,证明在脑缺血再灌注损伤中.细胞凋亡是神经元死亡的一种重要形式。

Abstract:In order to observe the pathologic injures of pyramidal cells of CA_1 region in the hippocampus 3 d after reperfusion following 10 min forebrain ischemia, bilateral carotid arteries occlusion model of old rats were used and the changes of ultrastructure of pyramidal cells of CA_1 region in the hippocampus were observed with electron microscope; meanwhile, we observed the changes of pyramidal cells in CA_1 region of the hippocampus through Tunel (Terminal-deoxynucleotidyl transferase-mediated dUTP-biotin nick and labeling) staining. The results showed: Compared with the sham-group,the ultrastructure of pyramidal cells of CA_1 region in the hippocampus 3 d after reperfusion following 10 min forebrain ischemia showed apoptosis-like changes.it has been tested that Apoptosis may be an important form in the course of cerebral ischemia-reperfusion(IR) injury.
