山茱萸多糖提取的两次优化工艺研究

摘    要:目的]优化提取山茱萸多糖的工艺参数。方法]在山茱萸多糖提取时间、提取温度和料液比3个因素最佳配合的基础上,选取酒精用量、沉淀时间2个因素进行二次回归正交组合设计试验,用响应面法对山茱萸多糖提取工艺参数进行了两次优化。结果]建立的山茱萸多糖提取产量的二次多项数学模型具有显著性(P<0.05)。失拟性检验结果表明,失拟不显著,回归模型与实际情况拟合得很好,从预测值与实测值的对比来看,也说明回归模型与实际情况拟合得很好。对回归方程求导可得到极值点,在酒精用量为6.52倍,沉淀时间59.7 h的条件下,山茱萸多糖提取最大预测值为35.22 g/100 ml,实际值为35.17 g/100 ml,两者基本相符。结论]利用优化工艺参数提取山茱萸多糖时具有最大的提取产量。

Abstract:Objective] The purpose was to optimize the technology parameters of polysaccharide extraction from Fructus Corni(F.Corni).Method] On the best coordination basis of 3 factors of extracting time,extracting temperature and solid-liquid ratio,the orthogonal combination of design of quadratic regression was tested by selecting 2 factors of alcohol amount and precipitation time,and optimization on technology parameters of polysaccharide extraction from F.Corni was conducted for two times.Result] The mathematical model of quadratic polynomial that built up for extracting yield of polysaccharide from F.Corni had significance(P<0.05).The test result for lack of fit indicated that lack of fit was not obvious,the fitting between regression model and actual conditions was very good,which also showed very nice from the comparison of predictive value with measured value.The extreme point could be got from derivation of regression equation.Under the conditions of alcohol amount as 6.52 times and precipitation time as 59.7 h,the maximal predictive value of polysaccharide extraction from F.Corni was 35.22 g/100 ml,which was basically in consistent with the measured value of 35.15 g/100 ml.Conclusion] The maximal extraction yield of polysaccharide from F.Corni would be obtained by using optimized extraction technology parameters.
