黄瓜CsGASA4基因克隆及生物信息学分析

摘    要:选取霜霉病与棒孢叶斑病两种病害胁迫后,在抗病品种中上调表达、感病品种中下调表达的基因克隆。通过生物信息学分析,初步开展该基因编码蛋白理化性质分析、亲疏水性预测、亚细胞定位预测、蛋白质结构预测。结果表明,该基因编码区全长315 bp,编码104个氨基酸,编码蛋白属于GASA super-family,具有跨膜结构,存在明显信号肽。系统进化树显示与大马士革玫瑰亲缘关系最近,同源性最高。根据其编码蛋白将该基因命名为Cs GASA4,亚细胞定位于分泌系统囊泡中。研究为进一步探究该基因功能及挖掘黄瓜双抗基因奠定基础。

Abstract:After stressing the two diseases of downy mildew and clonospora leaf spot, the gene which was up-regulated in resistant varieties and down-regulated in susceptible varieties was cloned. Through bioinformatics analysis, the protein encoded by the gene was initially analyzed for physicochemical properties, hydrophobicity prediction, subcellular localization prediction, and protein structure prediction. The results showed that the coding region of the gene was 315 bp in length and encoded 104 amino acids. The encoded protein belonged to GASA super-family, which had a transmembrane structure and a distinct signal peptide. The phylogenetic tree showed that the phylogenetic relationship with Damascus rose was closest and the homologywas the highest. Based on its encoded protein, the gene was named CsGASA4 and subcellular localization was in the vesicles of the secretory system. It laid the foundation of further exploring the function of the gene and digging out the double resistance genes of cucumber.
