中药复方抗鸡新城疫病毒作用研究

目的]探讨中药复方抗鸡新城疫病毒（NDV）的作用机理,为临床合理使用抗病毒性药物提供科学的理论依据。方法]以白花蛇舌草、金银花、黄芪和甘草组成中药复方,制备成中药复方流浸膏并配制成不同浓度梯度的药液;测定中药复方对鸡成纤维细胞（CEF）最大无毒浓度,并测定NDV在CEF上半数感染力（TCID50）;设中药复方不同浓度组,正常细胞和NDV感染细胞对照组,采用3种不同的作用方式作用于各组以检测中药复方抗NDV活性,采用四甲基偶氮唑蓝微量酶反应比色法（MTT法）测定OD值,以治疗指数为评价指标。结果]中药复方对CEF的最大无毒浓度为2-6g/ml,NDV对CEF的TCID50为10-7.3;第①种作用方式中药复方的最小有效浓度是2-10g/ml,第②种作用方式为2-8g/ml,第③种作用方式为2-7g/ml。结论]第①种作用方式对病毒的灭杀作用最强,第②种作用方式次之,第③种对病毒的灭杀作用最弱。表明中药复方不仅具有细胞外直接杀灭病毒作用,而且还具有抑制病毒吸附、穿入、复制和成熟的某一环节,为研究中药抗病毒的作用机理及临床应用提供了可靠的试验依据。

Effects of Chinese Herbal Compound on the Newcastle Disease Virus

Objective]To explore the antiviral mechanisms of the Chinese herbal compound on Newcastle disease virus(NDV)and to provide a scientific basis for the reasonable usage of the antiviral drugs in clinic.Method]The Chinese herbal compound was composed of Hedyotis diffusa,Lonicera japonica Thunb,Radix astragali and Glycyrrhiza uralensis.Then it was prepared to the Chinese herbal compound fluid extract with different concentrations.Its maximum no toxicity concentration and the tissue culture infective dose(TCID50)of NDV were respectively determined.There were normal cell control group,infected cell control group and treatment groups(different concentrations of the Chinese herbal compound).Different three methods were used for detecting the antiviral activities of the Chinese herbal compound.The OD value were determined by MTT3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide]method,and the therapeutic indexes were calculated to evaluate the antiviral effects.Result]The maximum of no toxicity concentration of the Chinese herbal compound was 2-6 g/ml,and the TCID50 was 10-7.3.The minimum effective concentration of Chinese herbal compound using the first method was 2-10 g/ml,that of using the second method was 2-8 g/ml,and that of using third method was 2-7 g/ml.Conclusion]The first method obtained the best antiviral effects,followed by the second method and the last was the third method,which indicated that the Chinese herbal compound not only kill the virus directly,but also inhibit the a certain process of the virus adsorption,penetration,replication and maturing.These provide a scientific basis for the reasonable usage of the antiviral drugs in clinic.
