“冷俊”槭查尔酮异构酶基因(CHI)片段克隆及序列分析

摘    要:以槭树品种"冷俊"为试材,利用RT-PCR技术,根据GenBank中登录的相关物种查尔酮异构酶基因(CHI)的保守序列设计简并引物,提取其秋季变色期叶片中的总RNA并扩增出CHI基因的cDNA片段。测序结果表明,该片段长553 bp,编码183个氨基酸。序列分析表明,该片段与龙眼、橄榄、山茶、沙梨等的同源性在81%~90%以上,证明已成功克隆到槭树变色期叶片CHI基因的cDNA片段,在GenBank中登录号为KC121346。

Abstract:The degenerate PCR primers were designed based on the conserved domain of some chalcone isomerase(CHI) genes in GenBank.The aimed cDNA fragment was amplified from the total RNA isolated from autumn color leaves of maple cultivar Marmo by RT-PCR.The sequencing results showed that the cDNA fragment was 553 bp encoding 183 amino acids.Sequence analysis indicated that the amino acid sequence shared 81%~90% homology to that of CHI genes from Dimocarpus longan,Canarium album,Camellia nitidissima and Pyrus pyrifolia.It confirmed that the cDNA fragment cloned in this study was from CHI gene of maple leaves.The accession number of this gene in GenBank was KC121346.
